چکیده

Estimates of caged-release based on photolysis of caged-fluorescein In order to provide estimates on the concentration of PtdIns(4,5)P2 or Ins(1,4,5)P3 released upon photolysis, we quantified the apparent efficacy of caged fluorescein photolysis in the first 40-50 μm of the pollen tube; this probe becomes fluorescent upon photorelease and the light emitted from loaded cells can be compared to known concentrations of the fluorophore from in vitro solutions (Malhó and Trewavas, 1996). The graph below (Fig. S1) depicts the estimated intracellular concentration of caged-fluorescein (X-axis)(based on the fluorescence emitted by the fluorophore upon a 5 sec flash of UV light) in pollen tubes submitted to an osmotic shock in a medium containing variable concentrations of the caged-probe (Y axis).

Phosphoinositides and phosphatidic acid regulate pollen tube growth and reorientation through modulation of [Ca2+]c and membrane secretion.

The maintenance of a calcium gradient and vesicle secretion in the apex of pollen tubes is essential for growth. It is shown here that phosphatidylinositol-4,5-bisphosphate (PIP2) and D-myo-inositol-1,4,5-trisphosphate (IP3), together with phosphatidic acid (PA), play a vital role in the regulation of these processes. Changes in the intracellular concentration of both PIP2 and IP3 (induced by p...

Turnover of Phosphatidic Acid through Distinct Signaling Pathways Affects Multiple Aspects of Pollen Tube Growth in Tobacco

Phosphatidic acid (PA) is an important intermediate in membrane lipid metabolism that acts as a key component of signaling networks, regulating the spatio-temporal dynamics of the endomembrane system and the cytoskeleton. Using tobacco pollen tubes as a model, we addressed the signaling effects of PA by probing the functions of three most relevant enzymes that regulate the production and degrad...

Effects of Copper and Leadon Pollen Germination Traits in Almond Cultivars

There is minimal information about the effect of heavy metals such as lead and copper on pollen grains and pollen tubes of fruit trees. Fruit set of these plants are affected by different environmental, biological, physical and chemical factors. If one of these factors be abnormal, pollination, fertilization and fruit set and orchard yield will decrease. In polluted cities, this phenomenon is a...

Endo/exocytosis in the pollen tube apex is differentially regulated by Ca2+ and GTPases.

Pollen tube growth relies on an extremely fast delivery of new membrane and wall material to the apical region where growth takes place. Despite the obvious meaning of this fact, the mechanisms that control this process remain very much unknown. It has previously been shown that apical growth is regulated by cytosolic free calcium ([Ca(2+)](c)) so it was decided to test how changes in [Ca(2+)](...

The Rrop GTPase switch turns on polar growth in pollen.

Pollen-tube growth not only represents an essential stage of plant reproduction but also provides an attractive model for studying cell polarity and morphogenesis. For many years, pollen-tube growth has been known to require a tip-focused Ca2+ gradient and dynamic F actin, but the way that these are controlled remained a mystery until recently. Rop appears to be activated at growth sites by a t...
